Diazyme’s 25-Hydroxy (25-OH) Vitamin D Assay Kit is in a microplate based ELISA format. The assay uses a Vitamin D specific antibody that effectively recognizes both 25-hydroxy Vitamin D3 and 25-hydroxy Vitamin D2, therefore the measurement of the total Vitamin D level is calculated in the blood sample. The assay accuracy quantifies Total 25-Hydroxy Vitamin D from 8.3 to 143.6 ng/mL, and has an excellent correlation with LC-MS assay with r2 >0.930 and slope of 0.941. The test shows excellent precision on a Dynex automated microplate reader with the intra-assay %CV were between 2.2% and 9.2%. The total %CV were between 4.9% and 9.4% for samples of Vitamin D levels ranging from 23.3 ng/mL to 116.8ng/mL.
The assay uses the enzyme immuno-assay technology (EIA). It is based on the competition (for a 25-OH Vitamin D antibody) between a 25-OH Vitamin D conjugate coated on a microplate and the 25-OH Vitamin D content a serum sample. Vitamin D samples are first extracted then mixed, in the coated microplate wells, with reagent R1 (containing an antibody for 25-OH Vitamin D) which allows for the competition to proceed. After an incubation, the microplate is washed and reagent R2 (HRP-labeled secondary antibody) is added. Following a second incubation and a washing step, reagent R3 (TMB substrate) is added. After a final incubation step,
the reaction is stopped by adding the STOP solution and the colorimetric signal of the microplate is measured at 450 nm. The whole assay procedure takes about two hours.
